Whole-body restoration

Research objectives and plan

The full restoration goals

Restore natural tooth substance and form, gums, living attachment, sensation and stable position after temporary support; pursue durable youthful function throughout the body.

Limits of this result

These are research objectives. Photos constrain visible form but do not uniquely recover hidden tooth thickness or internal damage.

Next useful step

Keep every required tissue and function visible when choosing a narrower experiment.

Dental restoration

Illustrative model

Support location and crown contact change the model response

Executed 30 support/contact cases with force, moment and energy checks. Equal support-length loss gives different compliance; two manufactured support distributions share the whole global stiffness matrix while local forces differ.

Limits of this result

Every length, stiffness and load is illustrative. The rigid crown does not model restored enamel thickness, biological healing, human mobility thresholds or stable position after support withdrawal.

Next useful step

Qualify root/socket geometry and spatial deformation or structural measurements alongside global response before fitting a tissue-specific model.

Dental restoration

Published data reanalysis

Oral tissue thickness, resistance and tracer transport differ

For two reported buccal cell-line model conditions, thickness differs 7.543-fold, electrical resistance-area product 1.016-fold and standard FD20 apparent permeability 0.577-fold.

Limits of this result

These are aggregate summaries with reported SEM and replicate counts, not paired donor/insert effects. Commercial total thickness includes matrix. The narrowed noninfectious comparison does not establish native gingival restoration.

Next useful step

Link identity, insert and time to geometry, resistance, tracer transport and viability before judging a functional gingival seal.

Tissue delivery

Source audit with conditional calculations

Delivery signals depend on time and reporter history

Compared six printed mouse-study associations: 1-hour and 24-hour qPCR each against a fixed 24-hour reporter. Display-rounded differences stay positive; late organ accumulation can disagree with reporter location.

Limits of this result

No new fit, raw paired animal series, intended-cell fraction or repair outcome is supplied. Reporter area retains prior activity, and three image fields do not establish three independent animals. Rounding bounds are not confidence intervals.

Next useful step

Connect animal, intended-cell denominator, molecular cargo scope and activity history to correction and organ function on traceable units.

Dental restoration

Observational data reanalysis

Living-pulp records retain sensory and imaging discordance

Linked all five patients in a human pulp pilot. At 24 weeks, four had positive electric sensibility; three also had unambiguously absent periapical radiolucency. An adverse radiograph coexists with positive sensibility in another patient.

Limits of this result

This is an uncontrolled five-patient measured pattern, not a regeneration success rate. MRI signal is not blood flow, late responses stay at their later visit, and CBCT timing/unit conflicts remain unresolved.

Next useful step

Link directly qualified perfusion, sensory response, pulp-space integrity and infection status on the same tooth through longer follow-up.

Dental restoration

Published data reanalysis

Aged interface data still lack measured repair-depth linkage

Recomputed 200 deposited OCT positions after thermal cycling. Conditional block summaries reproduce all four published SDs; descriptive CHX contrasts point in opposite directions across adhesive modes.

Limits of this result

Source blocks lack tooth IDs. OCT enhanced-signal width is a proxy, thermal cycling is distinct from mechanical fatigue, and separate tensile records cannot be joined to nominal repair depth.

Next useful step

Measure actual depth and pre/post-challenge interface behavior on the same identified specimens while retaining adverse failure modes.

Genome repair

Observational data reanalysis

Clinical function does not identify edited-cell coverage

Reconstructed six reported clinical-summary groups, four descriptive contrasts and the short follow-up timeline from the original single-patient CPS1 base-editing report.

Limits of this result

One infant, unequal sampling windows, unknown specimen counts and continuing care prevent an isolated causal effect. Liver biopsy was deferred; corrected-cell fractions and any ordinary-aging coverage threshold remain unknown.

Next useful step

Require causal-lesion evidence, relevant-cell coverage and durable organ function for each repair target; keep ordinary-aging target identification separate.

Dental restoration

Published data reanalysis

Dental bulk resistance and host attachment need separate measurements

Reanalyzed 14 listed crack-resistance curves, three contact-fatigue traces and three brushing-roughness summary pairs from the HEA workbook. Roughness worsens in all three material summaries.

Limits of this result

Listed traces and serial points do not identify independent specimens. Bulk response and the separate delamination traces do not link actual repair thickness to host-interface fatigue or clinical durability.

Next useful step

Connect measured regional thickness, host-junction behavior and retained-tooth damage under repeated loading on the same physical specimen.

Dental restoration

Published data reanalysis

Construct contraction is distinct from native ligament mechanics

Recomputed 777 micrometers of construct shortening and all six native-human tensile summary rows. Native summary moduli change across coupled force and loading-rate regimes.

Limits of this result

Force and shortening in the construct are algebraically linked. Active contraction and passive modulus cannot establish equivalent tissue function, donor-age effects, regenerated bilateral attachment or stable tooth position.

Next useful step

Link donor identity, construct geometry, both mineralized insertion interfaces, passive response and measured recovery before testing age-by-niche effects.

Compound delivery

Source audit with conditional calculations

MOTS-c persistence claims depend on what the assay measures

Qualified a primary ELISA/LC-MS disagreement and the K14Q steady-state comparison, preserving its variant-specific plasma calibration. None of the four bounded source contexts qualifies as administered-parent concentration-time PK.

Limits of this result

Signal-to-detection-limit scale separation is not true concentration or universal assay failure. One steady-state immunoreactive measurement does not identify intact-parent half-life or oral availability; donor-linked repair data remain unavailable.

Next useful step

Resolve intact administered parent, endogenous background and products, then connect exposure with the intended donor-stratified repair function.

Dental restoration

Published data reanalysis

48 teeth, with unresolved specimen identities

Analyzed 384 optical-margin rows and 48 fracture records. Thirty-seven teeth have internally consistent links; adverse joint-outcome bounds retain all 48.

Limits of this result

Eleven teeth have identity uncertainty. Deposited low load and blank margin fields stay unchanged. Preparation depth is nominal; these are extracted restored teeth, not regenerated tissue.

Next useful step

Add measured regional repair thickness and valid same-specimen histories while preserving the remaining tooth under loading.

Aging and youthful function

Observational data reanalysis

Age support changes the gingival trend

The full-cohort conflict spans nearly every fixed ECM gene. Restricting the bulk cohort to younger ages reverses its direction; shared-decade analysis preserves the earlier opposition.

Limits of this result

The common-span restriction removes every bulk donor over 45. These subsets are not matched-age or matched-donor validation, and expression is not restored function.

Next useful step

Seek matched-donor bulk/cell-resolved measurements with qualified sampling sites and independent functional outcomes.

Compound selectivity

Published data reanalysis

Selective signaling has not established reproductive protection

Original paired data reproduce DL001 selectivity: 433.38 as the mean of paired ratios, versus 350.24 as the ratio of means. The apparent discrepancy is resolved.

Limits of this result

Different assay states, unequal tissue responses and unmatched blood contexts remain. There is no direct DL001 reproductive-protection comparison or established healthy-human testicular-volume inference.

Next useful step

Specify useful function to retain and require same-context relevant-cell exposure plus direct reproductive outcomes.

Dental restoration

Published data reanalysis

What the measured dental material data show

Recomputed 75 stored source positions. Apparent toughness improves over acid-etched enamel but remains below the native mean; enamel wear directions are unfavorable.

Limits of this result

Source positions are not independent donors. Actual repair-depth and interface measurements are not linked to these specimen outcomes.

Next useful step

Find specimen-linked depth, crack, leakage and fatigue observations.

Dental restoration

Analysis software with synthetic examples

Keep dental depth and failures connected

The analysis preserves specimen/tooth/donor nesting, missing leakage and censored fatigue outcomes. The linked examples are synthetic.

Limits of this result

The real 75-position source extract supplies zero identifiable linked specimens. Software examples are not regenerated tissue or material performance measurements.

Next useful step

Apply the bridge to an eligible registered specimen series while keeping unknown targets and missing outcomes visible.

Aging and youthful function

Observational data reanalysis

Cell composition does not yet explain the age disagreement

Structural-ECM age slopes stay positive in observed and common-RNA partial mixtures, opposing the earlier bulk-tissue direction.

Limits of this result

The retained groups cover 99.55% of captured cells, but omitted RNA is unknown. This is not reconstructed whole tissue or a biological-age clock.

Next useful step

Read the follow-up gene/age-support challenge, then seek matched-donor bulk and cell-resolved measurements.

Compound delivery

Source audit with conditional calculations

SS-31: intact exposure and material accounting

Separated intact parent from metabolites, reproduced 21 cellular summaries and bounded oral material needs without assigning oral availability.

Limits of this result

Useful cellular access, oral availability and actual manufacturing cost remain unknown. Repeated source values across endpoints are retained as unresolved dependence.

Next useful step

Measure native-parent cellular exposure/function and released-parent process recovery before ranking modifications or delivery approaches.

Caffeine reversal

Illustrative model

Caffeine capture needs capacity as well as binding

A reversible, finite-capacity model uses published buffer affinities to test capture and competition; equimolar host does not imply complete capture.

Limits of this result

Chosen buffer concentrations are not human exposures or doses. Paraxanthine capture, oral delivery, brain exchange and sleep recovery are unresolved.

Next useful step

Qualify free-guest binding and intact accessible host in a physiological matrix before interpreting systemic capture.

Organoids and stem cells

Published data reanalysis

Relative spheroid response is not absolute function

Reconstructed 48 paired insulin observations. The flow/vascular-bed condition has the highest mean relative stimulation index, but not the highest absolute stimulated output.

Limits of this result

These are source cell-line spheroids, not patient grafts. Donor, lot and circuit crosswalks are unavailable; the pairs cannot be counted as independent people.

Next useful step

Link perfusion, identity, viability and function on the same constructs with their actual unit hierarchy.

Organoids and genome repair

Illustrative model

Separate repair endpoints do not establish joint success

The joint-endpoint audit shows why separate favorable measurements cannot establish how many units satisfy every required repair condition.

Limits of this result

The illustrative endpoint counts are synthetic. Allele, cell and tissue-area denominators cannot be silently combined.

Next useful step

Obtain joint observations with comparable denominators, unit IDs and the relevant functional outcome.
